The damage to nerve fibers was established in patients with endometriosis with severe pain. These changes correlate with neuronal hypertrophy and increased invasion of nerve fibers. The PubMed database was searched using relevant keywords to identify studies of invasion and damage to nerve fibers in endometriosis. The developed pathogenetic model of tissue innervation in endometriosis showed the presence of interaction of nerves and macrophages, indirect neurogenesis, the role of growth synchronization and differentiation between nerve fibers and microvessels, the contribution of microenvironment inflammation and nitrosative stress. Unnormal microenvironment inflammation and nitrosative stress are sources of excessive formation of neuropeptides, neurotrophins, angiogenic growth factors, which is facilitated by the activation of macrophages. Violation of the synchronization of growth and differentiation between nerve fibers and microvessels enhances indirect neurogenesis and the occurrence of chronic pain. The model for the development of pain in endometriosis allows to select a number of omix markers for diagnosing pain in endometriosis and to search for new therapeutic effects for treatment.
