Molecular mechanisms of neuroimmune interactions, e.g. the role of non-specific immunity in chronic pain, are paid a special attention. Toll-like receptors (TLRs), as a part of pattern-recognizing receptor group, are considered among the components of natural immunity response. Among the TLRs ligands, there are pathogen-associated molecular patterns (exogenic pathogens) and damage-associated molecular patterns (DAMPs), mainly expressed due to different stress reactions. Some neuropathic and dysfunctional mechanisms of chronic pain are realized via pathological activation of ion channels by pro-inflammatory mediators, as well as via aseptic inflammation following TLR activation. Further research of TLR role in chronic pain is a prospective direction in development of target medications for chronic pain.
