Thrombosis is a venous or arterial vascular that can sometimes cause fatal outcomes. Development of effective pharmacological agents for antithrombotic therapy is still an urgent scientific and practical objective.

OBJECTIVE

To study the antithrombotic effects of blue mussel (Mytilus edulis) hydrolysate preparation.

MATERIAL AND METHODS

Biochemical methods were used to analyze the effect of blue mussel hydrolysate on enzymatic activity of key serine proteases in clotting cascade. We assessed the structure of actin cytoskeleton and endothelial monolayer morphology in vitro to investigate the effect of this drug on biological activity of thrombin. Anticoagulant effect of hydrolysate was investigated in a model of posterior vena cava thrombosis in rats.

RESULTS

The drug inhibits serine proteinases of clotting cascade (FIIa, FXa, FXIa, FXIIa), in particular, enzymatic activity of thrombin (FIIa) against fibrinogen and synthetic substrates Tos-Gly-Pro-Arg-pNA and Ala-Ala-Arg-pNA. Anti-thrombin activity of undiluted drug was about 6.5 AT-U/ml. Hydrolysate inhibited the ability of thrombin to cause actin cytoskeleton remodeling and endothelial monolayer disintegration. Subcutaneous injections significantly changed thrombus structure and reduced the probability of clotting in a model of posterior vena cava thrombosis in rats.

CONCLUSION

M. edulis hydrolysate may be a basis for development of new drugs for therapy of hypercoagulable conditions.
