OBJECTIVE

To evaluate neutrophils extracellular traps (NETs) formation in a patient with ulcerative colitis (UC).

MATERIAL AND METHODS

NETs formation was studied on neutrophils isolated from the whole blood of a UC patient on a ficol-verografin gradient. We compared NETs formation in UC patient and healthy volunteers (controls) in intact neutrophils and after 30 minutes of probiotics stimulation.

RESULTS

Neutrophils in a patient with UC demonstrated a more pronounced ability to NETs formation both intact and after stimulation. NETs formed in a UC patient after probiotics stimulation had significant network size, exceeding the neutrophil size by 10—18 times, while NETs in controls were only 4—8 times larger. The NETs of UC patients demonstrated a more pronounced ability to capture bacilliform bacteria, with 21 bacilli captured, compared to controls Me 7.5 (LQ 4.75; HQ 10.5). Except for evaluating the NETs formation, UC activity in a patient was assessed by colonoscopy, histology, complete blood count, C-reactive protein, and fecal calprotectin.

CONCLUSION

NETs formation in UC patient was more pronounced than in healthy controls. Proinflammatory proteins, cytokines, and other inflammatory mediators in the blood of UC patients may change the composition and density of receptors on the neutrophil membrane and trigger receptor-mediated NETosis.
