The authors studied the efficacy and tolerability of the Russian generic agent EXORUM (oxaliplatin) in combination with calcium folinate and 5-fluoruracil (an mFOLOX6 regimen) in the first-line chemotherapy (CT) of disseminated colon cancer (DCC). In October 2008 to January 2012, the trial enrolled 40 patients (mean aged 60 years). A total of 303 courses of CT were performed. Its objective effect was recorded in 47.5% of the patients; disease stabilization of more than 8 weeks was noted in 30%. The median time to progression was 6.4 months; the median overall survival was 16.4 months. Six patients were found to have grade III-IV hematological toxicity, including a febrile neutropenic episode. Grade I, II, and III sensory neuropathy was observed in 32 (80%), 10 (25%), and 2 (5%) patients, respectively. When the first signs of neuropathy appeared, pre- and posttreatments with Ca2+ and Mg2+ preparations were performed in all patients. Thus, EXORUM demonstrated the efficacy and safety comparable to those shown in large-scale randomized trials.
