THE PURPOSE OF THE STUDY

Was to investigate the expression pattern of toll-like receptors, the immunophenotype of lymphocytes, the cytokine profile of patients with chronic spontaneous urticaria (CSU).

MATERIAL AND METHODS

In 134 patients with CSU, TLR expression 2, 3, 4, 9 was studied on mononuclear leukocytes of peripheral blood (MLPB) using monoclonal antibodies («Caltag Laboratories», USA) using an FC-500 flow cytometer («Beckman Culter», USA). The immunophenotype of lymphocytes (CD3+/CD45+, CD3+/CD4+, CD3+/CD8+, CD16+/CD56+, CD45+/CD25+, CD72+, CD45+/CD95+) was studied by flow cytometry method. The cytokine profile (IFN-γ, IL 1β, 2, 4, 5, 10) of serum/plasma of blood was studied by enzyme-linked immunosorbent assay method.

RESULTS

In patients with CSU were revealed, low expression levels of TLR 2, 3, 4, 9, a decrease in the content of CD3/CD45, CD3/CD4, an increase in the level of CD45/CD25, CD45/CD95 on lymphocytes, an increased production of pro-inflammatory IL-1β, anti-inflammatory IL 4 and 5 against normal levels of production of IL 2, 10 and IFN-γ.

CONCLUSIONS

A decrease in the number of MLPB expressing TLRs 2 and 4 indicates a possible relationship between innate immunity and the etiopathogenesis of CSU. An increase in the expression of IL-4 in serum indicates the activation of T-lymphocytes by the Th2 pathway in patients with CSU.
