Mesenchymal stromal cells (MSCs) are a heterogeneous population of progenitor cells presenting in perivascular compartment of most tissues and performing important regulatory functions. MSCs regulate angiogenesis, neurogenesis, immune reactions and reorganization of extracellular matrix due to secretion of various biologically active molecules. This makes them important for cell therapy and tissue engineering. MSCs of bone marrow, adipose tissue and perinatal tissues are the most studied. However, there are few data on cardiac MSCs and modulation of their paracrine function to enhance regenerative properties.
To study secretome of MSCs derived from human heart and cultured under 3D conditions (as spheroids); to evaluate its angiogenic properties. We found that human cardiac MSCs contain proangiogenic factors (VEGF, bFGF, IL8, Rantes, etc.) stimulating migration of endothelial cells in vitro and angiogenesis in vivo.
These results create a basis for further studies devoted to development of new cell-free products based on human cardiac MSC secretome for the treatment of ischemic cardiac diseases.
