INTRODUCTION
The protein ANXA2, encoded by the ANXA2 gene, performs a number of biological functions that are primarily related to cellular transport. Disturbances of functional activity gene ANXA2 have been detected in tumor-diseases, but, for this gene we have previously shown increased expression of the Anxa2 gene at the mRNA level in the early stages of neurodegeneration processes. In this regard, the most interesting is studying decrease of expression gene anxa2a to the functioning nervous system on the model D. rerio with used morpholino oligonucleotides (morpholino). In this manuscript we are studying the influence of morpholino to gene anxa2a on the embryo development of Danio rerio.
MATHERIAL AND METHODS
The study was conducted with used Danio rerio line AB, whose embryos were injected of morpholino oligonucleotides to gene anxa2a. The estimate injected morpholino oligonucleotides on the embryo development is effected through by analysis change of phenotype to 2 and 4 day post fertilization (dpf).
RESULTS
Injection of experimental morpholino experimental morpholinos targeting the start-codon region and 5’UTR leads to an increase in the percentage of deformations compared with the Co-In and Co-Mo-D groups by 4 dpf. The deformations analyzed by 4 dpf were observed in control and experimental groups. In addition, we analyzed an earlier time point, at 2 dpf. Phenotype changes at this time point were observed only in the experimental groups.
CONCLUSION
Comparative data analysis, obtained from injected morpholino, targeted to start-codon and 5’UTR of gene anxa2a, showed that both variants may be used for future study. Also, we have shown that preferably, to conduct analysis of expression in 2 dpf. The effects detected at 2 dpf, to all appearance, to be specific, and the presence of a hindbrain ventricular deformity suggests that altered expression of the anxa2a gene may lead to nervous system dysfunction.
